Today you will be digesting mycobacteriophage Mozy DNA with restriction enzymes (BamHI), (ClaI), and (HindIII). Without knowing anything about the DNA sequence of Mozy, it will be possible for us to determine if this phage is closely related to sequenced mycobacteriophages by comparing the Mozy pattern of restriction fragments with patterns that you will deduce from sequenced mycobacteriophages.

Since there are no published gels for mycobacteriophage restriction digests, we will use a computer program to predict the sizes of restriction fragments. Then we can draw the gels by hand. We will then compare the gel we run in class with the predictions to see which (if any) of the published phages is most closely related to Mozy. Use the semi-log graph paper distributed in class, the table below, GenBank, and Webcutter to complete your assignment. Your completed drawing should include 10 lanes. The order of the lanes is up to you, but label them appropriately:
